Tesamorelin/Ipamorelin Blend – 10mg/3mg
For Research Use Only. Not for human or veterinary use.
Tesamorelin/Ipamorelin is a research peptide blend combining two distinct growth hormone secretagogues that act through different signaling pathways. Tesamorelin is a synthetic 44-amino-acid analog of human growth hormone-releasing hormone/factor (GHRH/GRF), while ipamorelin is a synthetic pentapeptide growth hormone secretagogue originally characterized for selective stimulation of growth hormone (GH) release.
Tesamorelin has been extensively investigated in controlled human studies involving GH–IGF-1 signaling, visceral adipose tissue, lipid metabolism, and body-composition biology. Ipamorelin has been investigated primarily for its growth-hormone-releasing pharmacology, receptor selectivity, and pharmacokinetic/pharmacodynamic characteristics. Published research supporting the individual compounds should not be interpreted as establishing the efficacy or safety of the specific 10mg/3mg combination.
Mechanism of Action
Tesamorelin acts as an agonist of the growth hormone-releasing factor receptor. FDA pharmacology data indicate that tesamorelin binds and stimulates human GRF receptors with potency similar to endogenous GRF. Activation of this pathway stimulates pituitary synthesis and pulsatile release of endogenous growth hormone, which can subsequently increase circulating insulin-like growth factor-1 (IGF-1).
Ipamorelin works through a separate growth hormone secretagogue pathway. The original pharmacological characterization identified ipamorelin as the pentapeptide Aib-His-D-2-Nal-D-Phe-Lys-NH2 and demonstrated GH release through a GHRP-type receptor. More recent FDA evaluation describes its primary nonclinical mechanism as ghrelin-receptor agonism. In experimental animal studies, ipamorelin produced potent GH release while showing greater selectivity for GH than GHRP-2 or GHRP-6 with respect to ACTH and cortisol release.
Human pharmacokinetic/pharmacodynamic research has also demonstrated dose-related GH responses following ipamorelin exposure. In a healthy-volunteer study, ipamorelin generated an episodic GH response with a peak approximately 0.67 hours after infusion and had a reported terminal half-life of approximately two hours.
Because GHRH-receptor agonists and GHRP/ghrelin-receptor agonists engage different components of the GH secretory system, dual-pathway stimulation is an established area of endocrine research. Human experiments combining GHRH with other GHRPs, particularly GHRP-6 and GHRP-2, have produced enhanced GH responses compared with stimulation of either pathway alone. These studies provide a mechanistic basis for investigating complementary GHRH and GHRP signaling, but they did not study the specific tesamorelin/ipamorelin 10mg/3mg blend, so equivalent interaction or clinical effects cannot be assumed.
Research Applications
Studies involving the individual peptides or their respective signaling classes have explored several areas of experimental research:
Growth Hormone Secretory Dynamics
GH–IGF-1 Axis Research
GHRH/GRF Receptor Signaling Research
Growth Hormone Secretagogue and Ghrelin-Receptor Research
Dual Secretagogue Pathway Interaction Studies
Visceral Adipose Tissue Biology
Body Composition and Metabolic Research
Lipid and Adipose Tissue Research
Peptide Pharmacokinetic/Pharmacodynamic Studies
Endocrine Receptor Selectivity Research
Tesamorelin in particular has demonstrated reductions in visceral adipose tissue in randomized controlled studies of adults with HIV-associated abdominal fat accumulation. In one 412-participant trial, visceral adipose tissue decreased by 15.2% over 26 weeks in the tesamorelin group while increasing by 5.0% with placebo; later studies also examined liver-fat biology. These findings are specific to tesamorelin under the studied clinical conditions and do not establish the same outcomes for ipamorelin or the combined research formulation.
Tesamorelin is present in FDA-approved prescription products for the reduction of excess abdominal fat in HIV-infected adults with lipodystrophy, whereas FDA has stated that there are no FDA-approved drug products containing ipamorelin or ipamorelin acetate. The specific tesamorelin/ipamorelin blend described here is therefore distinct from an FDA-approved tesamorelin drug product.
