Thymosin Alpha 1 Peptide

For Research Use Only. Not for human or veterinary use.

Thymosin Alpha 1 (Tα1), also known in its synthetic form as thymalfasin, is an N-terminally acetylated 28-amino-acid peptide originally characterized from thymosin fraction 5. It is a naturally occurring fragment associated with prothymosin alpha and has been studied primarily for its immunomodulatory activity.

Thymosin Alpha 1 is primarily investigated in immune-system research for its potential influence on dendritic cells, T-lymphocyte function, innate immune signaling, antigen presentation, cytokine regulation, and host-defense responses. Research has also examined the peptide in viral, fungal, inflammatory, vaccine-response, critical-illness, and tumor-immunology models.

Mechanism of Action

Research suggests Thymosin Alpha 1 may influence both innate and adaptive immune responses through several interconnected signaling pathways rather than through one conclusively established receptor mechanism. In specific dendritic-cell and infection models, Tα1 activity has been associated with Toll-like receptor signaling, particularly TLR2 and TLR9, followed by MyD88-dependent activation of pathways involving p38 MAPK, NF-κB, and interferon regulatory factor 7.

Experimental studies using human monocyte-derived dendritic cells have reported increased expression of antigen-presentation and costimulatory markers, including MHC class I, MHC class II, CD40, and CD80. These observations have made Tα1 a subject of interest in research involving dendritic-cell maturation, antigen presentation, and the coordination of innate and adaptive immune responses.

Thymosin Alpha 1 has also been investigated for its potential effects on T-cell differentiation and activation, T-helper type 1 immune responses, natural-killer-cell activity, macrophage function, and cytokine production. Its observed activity can vary according to the immune cell type, biological stimulus, and experimental model, so it is generally classified as an immunomodulatory peptide rather than solely as an immune stimulant.
